This is all the info I got from doing the beta.
The info may be subject to change, but for a general idea, here is this guide.
And with new info from Grant's post, I'm going to have to fact check a few things.

On Lava

Ancient Stronghold Shrines

Capture these and hold them for a specific time specified (2 hours for now) and you are ruler of the realm and can spawn cool prizes for you and/or the realm. You become ruler and even get to pick a message for the realm, have your name in yellow, AND get your avatar and name on blast in the event page.
TO lead a rally to the Stronghold Shrine, you need to BE on the Lava. You get no biome bonuses on the lava. To join a rally to the Stronghold Shrine, you need to be on the Lava OR on one of the Biome Isles with the mini shrines and their cores, you can not join a rally to the Stronghold Shrine from the Mainland.

On the Biome Shrine Lands

Cores

On the Biome Shrine lands, you’ll find a Grasslands, Badlands and a Swamp Core. You and your clanmates can capture these to help your clan to get bonuses to their Dragons and Attack Powers for the Stronghold. These really help you to capture and hold the Ancient Stronghold Shrine. Your clan can only Hold 1 of these at a time, and in order to lead a rally to these, you need to be on the specific island that holds this core. In order to JOIN a rally to the Core, you need to be on the Lava on one of the Biome Isles, (or on the Mainland).

The Cores give you a boost for your attacks on the Stronghold as well as a bonus for your dragons specific to the biome the core is on. This helps you hold the Ancient Stronghold Shrine and Mini Shrines.
For example, this grasslands core gives your grasslands dragons a boost when you use them on grasslands. You also get an attack boost when you attack the Ancient Stronghold.

Mini Shrines
​

Also on the biome specific lands that hold the Cores, you’ll find Mini Shrines. The North Biome Shrine is to the north of the Core, the South Biome Shrine is to the South of the Core, the East Biome Shrine is to the East of the Core, and you guessed it, the West Biome Shrine is to the West of the Core. Each Mini Shrine is named for the biome they are on, and the location in relation to their Core. So in Grasslands, you’ll have the Grasslands Core and surrounding it will be the North Grasslands Shrine, South Grasslands Shrine, East Grasslands Shrine and West Grasslands Shrine. In order to lead a rally to a shrine, you will have to be on that island. (You need to be on the Grasslands Isle to lead a rally to the Grasslands Shrines or core, but you can join it from the Lava from any of the Biome Isles, or from the Mainland. Capturing these shrines will give you and your clan boosts in that biome. If you/your clanmates capture grasslands shrines, you will find it easier to hold the Grasslands Core because your grasslands marches are stronger with those boosts. Your other biome marches won't be stronger, and your marches on Lava won't be affected since it is Lava, not Grasslands, Badlands or Swamp.

March Limitations On Specific Lands

Lava​

If you are on the lava, you can’t shield.

Marches To Mainland:
While on the lava you can not do anything on the Mainland except send resources to your clanmates. You can’t scout people on the mainland, you can’t attack those on the mainland, gather resources, attack monsters etc.

Marches To Biome Isles:
You also can’t gather rss or attack monsters or cities that are on the Biome Isles. You CAN scout people who are on the Biome Isles, you can defend anyone who is on the biome isles, and you can join rallies to shrines and cores that are on the isles, and you can send resources to your clannies who are on the Biome Isles (and the mainland) but you can’t start an Attack march to the biome isles. No attacks, no PvE, no PvP, no Starting a PvP Rally.

Tested: If a Rally is Started by a clanmate who is on the Biome Isles, you CAN join from the Lava.
If a Rally is Started by a clanmate who is on the Mainland and you are on lava, you Can Not join because you can not reach the Mainland from the Lava.

If while on the Lava, someone attacks you AND WINS the attack against you, your City gets teleported to a random location on the map and you’re given a free teleport to port to a location of your choice.

Biome Isles

Marches To Biome Isles:
While on the Biome Isles, you can Lead a rally to the Core or Shrine of your specific biome, but not of other biomes. You Can join rallies to any biome Cores and Shrines.

Marches To Lava:
You Can scout and send resources to and reinforce your clanmates who are on Lava, you CAN NOT send Attack marches to the Lava. No PvP, no Rallies on Cities, no gathering.
You Can join rallies to the Ancient Strongholds too

Marches To Mainland:
You Can also scout the mainland, you can attack cities on the mainland, do PvE, PvP, Rallies, Gather RSS, and Send RSS to clannies and reinforce members who are on the Mainland from the Biome Isles.
Grant's new and updated restrictions say you can do all types of marches from the biome isles to the mainland.

Mainland​

Marches To Lava:
When on the Mainland, you can’t lead or join rallies to the Ancient Stronghold Shrine. You can not do anything to anyone who’s on the lava except send resources.

Marches To Mainland:
You CAN do everything you always could to anyone on the mainland since you’re on the mainland.

Marches To Biome Isles:
When on the Mainland, you can’t lead rallies to the Biome Isles Core or Mini Shrines, but you can join them.
You can also scout people on the Biome Isles, send attack marches PvE, PvP, Rallies to the Biome Isles. You can Gather RSS, and send RSS to clannies. You can also send Reinforcements to Cities on the Biome Isles.

To tell if you're on the Mainland or on Lava or on one of the Shrine islands, the easiest way is to look at the map. ​

This sand divides the Mainland from the islands.


Upgrade your Waritorium
to increase your marches on the
Shrines and Strongholds.

You can find those caps in the Player Profile under the "Clan" section.​

Thank you. I am one of the players who = too small to participate in the event. I only participated in it in a beta test of it, but not in the real game... It's not meant for me. I am kind of disappointed too, but c'est la vie. I can't see myself being one of those top players who can participate in Shrines any time soon.
